CR has a specific expertise of mammalian cells in culture and molecular cell biology dating back to her time as a student when she detected by DNA fingerprinting the induction of mutations by recombinogenic agents in an established cell line. She first became interested in the regulation of DNA precursors (dNTPs) in mammalian cells when she was an Erasmus fellow at Stockholm University. She first studied the enzymes involved in dNTP de novo synthesis and salvage during the cell cycle and then turned to the study of 5'-nucleotidases. In 1999 she discovered the first known deoxynucleotidase in mitochondria (mdN) which operates in an intramitochondrial substrate cycle regulating mitochondrial dTTP synthesis. She demonstrated that the susceptibility of a nucleotide analogue to be dephosphorylated by 5’nucleotidases is a predictive factor of the drug efficiency in anticancer or antiviral therapy. She studied the mechanisms that in mammalian cells keep the dNTP pools in balance both in the cytosol and in mitochondria. Over the last several years she has been working on the latest known dNTP catabolic enzyme, SAMHD1. She has recently demonstrated that SAMHD1 influences the dynamics of mitochondrial dNTP pools. She is presently analyzing the effects of SAMHD1 loss of function in fibroblasts from patients affected by the Aicardi Goutières syndrome.
